The British Heart Foundation (BHF) funded Yorkshire and Midlands Family Heart Studies were launched at the end of 1998 to investigate the genetic basis of coronary heart disease through the study of affected sibling pairs (ASPs). 

Since then, with the help of clinical colleagues and also the local media (newspaper, radio, TV) we have identified, interviewed, obtained blood and prepared DNA from over 800 pairs of siblings affected by premature coronary artery disease (myocardial infarction, coronary artery bypass surgery, percutaneous transluminal coronary angioplasty, proven stable / unstable angina up to age of 65yr). 

Detailed methodologies for case identification, screening, contact, counselling and validation have been developed and refined to permit extension to a second national phase with intended identification and study of a total of 2,400 ASPs and their surviving unaffected siblings. The latest automated fluorescence-based techniques are being used for large-scale genotyping (2 million genotypes) to create a 10cM genome-wide map. Databases have been developed and refined to permit extension to a second national phase with intended identification and study of a total of 2,500 ASPs and their surviving unaffected siblings. 

The collection of data and DNA from a further 3,725 unaffected siblings will prove extremely valuable for subsequent fine mapping of loci and for testing the candidacy of specific genes through intra familial association studies. MRC funding will permit provision of additional DNA as well as cryopreservation of lymphocytes, thereby providing a limitless national resource for the study of acute and chronic premature coronary artery disease.
